چکیده

A unique pentasaccharide fragment of heparin can enhance the reactivity of antithrombin with coagulation proteases factors IXa and Xa by 300to 600-fold through a conformational activation of the serpin, without having a significant effect on the reactivity of antithrombin with thrombin. In this study, it was hypothesized that differences in the structure of the autolysis loop of coagulation proteases (residues 143-154 in chymotrypsin numbering) may be responsible for their differential reactivity with the native and heparin-activated antithrombin. To test this hypothesis, the autolysis loops of both thrombin and the anticoagulant serine protease–activated protein C were replaced with the corresponding loop of factor Xa. Inhibition studies revealed that in contrast to the approximately 1.5-fold difference in the reactivity of thrombin with antithrombin in the absence and presence of pentasaccharide, the difference in reactivity was increased to approximately 37-fold for the mutant thrombin. In the case of the activated protein C mutant, similar to factor Xa, pentasaccharide accelerated the reaction 375-fold. These results suggest that structural differences in the autolysis loop of coagulation proteases play a key role in their differential reactivity with the native and heparin-activated conformations of antithrombin. Atherosclerosis: testing the water.

Acritical regulatory pathway of the blood coagulation cascade is provided by antithrombin, a serine protease inhibitor (serpin) that specifically shuts down the activity of factor Xa and thrombin.1 Much of what we know about the mechanisms of antithrombin function has come from studies using the cofactor heparin, that allosterically activates antithrombin for optimal presentation to the target ...

Intrinsic pathway of coagulation and arterial thrombosis.

Formation of a fibrin clot is mediated by a group of tightly regulated plasma proteases and cofactors. While this system is essential for minimizing blood loss from an injured blood vessel (hemostasis), it also contributes to pathologic fibrin formation and platelet activation that may occlude vessels (thrombosis). Many antithrombotic drugs target key elements of the plasma coagulation mechanis...

HEMOSTASIS, THROMBOSIS, AND VASCULAR BIOLOGY Human single chain antibodies against heparin: selection, characterization, and effect on coagulation

Heparin, located in mast cells and basophilic granulocytes, is widely used as an anticoagulant. It belongs to a class of linear polysaccharides called glycosaminoglycans (GAGs). Using phage display technology, we have selected 19 unique human antiheparin antibodies. Some antibodies react almost exclusively with heparin, others also react with the structurally related heparan sulfate, and some w...

Role of tissue factor in hemostasis, thrombosis, and vascular development.

Tissue factor (TF) is best known as the primary cellular initiator of blood coagulation. After vessel injury, the TF:FVIIa complex activates the coagulation protease cascade, which leads to fibrin deposition and activation of platelets. TF deficiency causes embryonic lethality in the mouse and there have been no reports of TF deficiency in humans. These results indicate that TF is essential for...

Thrombin generation in hemorrhage control and vascular occlusion.

Hippocrates and Aristotle recognized that blood clots outside the body, but it was not until 1720 that John Louis Petit recognized that control of hemorrhage after amputation was associated with the blood clotting process.1 One hundred thirty-eight years later, Rudolph Virchow formulated his postulates regarding clots in venous thrombosis.2 The association of blood clots with acute arterial occ...
